Research
This laboratory is currently characterizing and studying
the regulation of a family of carboxylesterases which
hydrolyze fatty acid esters of cholesterol, glycerol
and retinol in various tissues, including liver,
testis and brain. Interest is focused on a liver
isoenzyme which is the major cholesterol ester hydrolase
activity in the cytosolic compartment. Previous work
from this laboratory suggests that this enzyme plays
an important role in regulation of hepatic free cholesterol,
which is the substrate for bile acid synthesis, as
well as an essential constituent of membranes and
lipoproteins. Work in progress involves 1) measurement
of enzyme properties, activity, mass and mRNA levels
with conditions or perturbations known to influence
cholesterol metabolism, 2) characterization of the
promoter for the hepatic cholesteryl ester hydrolase
and 3) cloning and expression of various isoenzymes
to explore the physiological significance of minor
sequence differences.
